Dieses Projekt hat das Ziel, ein systembiologisches Verständnis der Biofilm-Hemmung durch kleine Moleküle zu erarbeiten. Auf dieser Basis sollen neue Wirkstoffe gegen Biofilme entwickelt werden, die nicht toxisch sind und daher in der Präventivmedizin eingesetzt werden können. Speziell werden wir uns zunächst auf den kürzlich entdeckten Biofilm-Hemmstoff Carolacton konzentrieren, einen Sekundärmetaboliten aus dem Myxobakterium Sorangium cellulosum, der die Biofilmbildung des Kariesbakteriums Streptococcus mutans in nanomolaren Konzentrationen hemmt. Die Untersuchung wird auf klinische Isolate von Streptococcus und auf andere Bakterien ausgeweitet, die Biofilme bilden und für Infektionen im menschlichen Körper verantwortlich sind. Eine Totalsynthese von Carolacton und seine Modifizierung auf Grund von Hypothesen wird durchgeführt um die Hemm-Aktivität zu verbessern, und um sie mit klinisch relevantem Material zu koppeln, z.B. Implantaten, Schläuchen, und Zahnfüllungen, die im Tiermodell getestet werden.
